Researchers unveil first artificial enzyme created by evolution in a test tube

Thursday, January 31, 2013

There's a wobbly new biochemical structure in Burckhard Seelig's lab at the University of Minnesota that may resemble what enzymes looked like billions of years ago, when life on earth began to evolve ? long before they became ingredients for new and improved products, from detergents to foods and fuels.

Seelig created the fledgling enzyme by using directed evolution in the laboratory. Working with colleague Gianluigi Veglia, graduate student Fa-An Chao, and other team members, he subsequently determined its structure, which made its debut December 9 as an advance online publication in Nature Chemical Biology. Lab tests show that the enzyme (a type of RNA ligase, which connects two RNA molecules) functions like natural enzymes although its structure looks very different and it is flexible rather than rigid. Seelig speculates the new protein resembles primordial enzymes, before their current structures evolved.

Seelig and Veglia are professors in the College of Biological Sciences, where Fa-An Chao is a graduate student. Both faculty members have appointments in the Department of Biochemistry, Molecular Biology and Biophysics and Seelig is member of the BioTechnology Institute.

While a handful of groups worldwide are developing artificial enzymes, they use rational design to construct the proteins on computers. Instead, the Seelig lab employs directed evolution. "To my knowledge, our enzyme is the only entirely artificial enzyme created in a test tube by simply following the principles of natural selection and evolution," he says.

Rational enzyme design relies on preconceived notions of what a new enzyme should look like and how it should function. In contrast, directed evolution involves producing a large quantity of candidate proteins and screening several generations to produce one with the desired function. With this approach, the outcome isn't limited by current knowledge of enzyme structure.

"Just as in nature, only the fittest survive after each successive generation," Seelig explains. The process continues until it produces an enzyme that efficiently catalyzes a desired biochemical reaction. In this case, the new enzyme joins two pieces of RNA together.

"It's kind of like giving typewriters to monkeys," he says. "One monkey and one typewriter won't produce anything clever. But if you have enough monkeys and typewriters, eventually one of them will write 'to be or not to be'." The lottery provides another analogy. "If you buy more tickets, you're more likely to win," Seelig says.

Like all proteins, the new RNA ligase enzyme is a chain of amino acids folded into a 3D structure, but the resemblance stops there. Natural enzymes, like all proteins, are made from alpha helices and beta strands. Seelig's artificial enzyme lacks those structures. Instead, it forms around two metal ions and is not rigid. "Compared to enzymes we know from nature, the artificial enzyme has a rather unusual structure and dynamics," Seelig says.

For decades, naturally occurring enzymes have been tweaked by industry to make industrial processes and products more effective. The ability to create enzymes from scratch using a natural process opens the door to a vast array of new products that provide business opportunities and improve quality of life without harmful environmental effects.

Going forward, Seelig plans to create enzymes with useful applications while he continues to explore the underlying basic science of enzyme structure and function, aiming to learn more about the origin of enzymes and how proteins evolve.

"Enzymes have always fascinated me," he says. "It's rewarding to do work that has practical applications yet provides the opportunity to better understand how life on earth evolved."

McCain: Immigration reform failure would cost GOP

(AP) ? In a warning to fellow Republicans, Sen. John McCain said Wednesday that failure to pass comprehensive immigration legislation could mean continued election losses for the GOP and cause Republican-friendly states like Arizona to fall to the Democrats.

The Arizona Republican, his party's former presidential nominee and one of eight senators to sign onto a bipartisan immigration reform framework this week, said failure to act means the trend of Hispanic defections from the GOP would continue.

Latino voters supported President Barack Obama over Republican Mitt Romney 71 percent to 27 percent in November, helping to ensure Obama's victory.

McCain said that Republicans have failed to understand the importance of immigration to Hispanic voters and that's cost the party at the ballot box.

"If you have a large bloc of Americans who believe you're trying to keep their ... fellow Hispanics down and deprive them of an opportunity, obviously that's going to have an effect," McCain said.

The Senate framework would provide a path to citizenship for the estimated 11 million illegal immigrants already in the country, secure the border, attempt to simplify legal immigration, and add requirements for employers to prevent hiring of illegal immigrants.

If no bill passes, McCain said, the forecast for Republicans is that "the trend will continue of lack of support from Hispanic voters, and also as you look at the demographics of states like mine that means that we will go from Republican to Democrat over time."

McCain spoke alongside Democratic Sen. Charles Schumer of New York, his partner on the immigration proposals, at a breakfast hosted by Politico.

Review: 'Revenge' filled with haunting prose

"Revenge: Eleven Dark Tales" (Picador), by Yoko Ogawa, translated by Stephen Snyder

They are the scenes of ordinary life: a mother stopping by the neighborhood bakery to purchase two strawberry shortcakes for her son's birthday, an aspiring writer toiling over a manuscript in a spare apartment, a young woman preparing dinner for her beau, a woman spying on her husband's mistress.

Yet in Yoko Ogawa's story collection, "Revenge: Eleven Dark Tales," those ordinary exteriors are merely brittle shells that crack open to reveal darkness, death and despair. Woven through the 11 interconnected tales is a thread of the grotesque, the macabre, the mournful.

The mother's errand turns out to be a paean to inconsolable loss. The writer emerges as an unhinged character that evokes both love and pity. The amorous young woman finds herself entwined in both a murder scene and a museum dedicated to torture.

Ogawa, a prolific author whose work has appeared in The New Yorker and Harper's Magazine, laces her stories with gruesome murders, exotic animals and peculiar events. Her language is both spare and searingly precise, crystallizing the details of everyday existence and capturing the unexpected shock of the bizarre.

In "Sewing for the Heart," for example, the narrator is a bag-maker who has been contracted to create a purse for a beating heart. The client is a cabaret singer who was born with the organ outside her chest. The narrator gasps in awe at the sight and utters an oddly erotic ode to the throbbing muscle: "What extraordinary, breathtaking beauty! Would it feel damp if I cupped it in my hands? Would the membrane rupture if I gave it a squeeze? ... I wanted to run my fingertips over each tiny bump and furrow, touch my lips to the veins, soft tissue on soft tissue, the pressure of her pulse against my skin."

Many of Ogawa's characters, including the bag-maker and the cabaret singer, reappear in other stories, as do details and events in ways that are sometimes incidental, sometimes integral to the plots. The effect is, as Ogawa describes the novel written by the unhinged author, an "icy current running under her words."

In these stories, ordinariness is not a mask hiding the morbid and the macabre. In many cases, the ordinary life itself, with its insistent drip of isolation, sameness, sadness and loss, is what pushes the characters to the edge of madness and vengeance.

As Ogawa writes in "Welcome to the Museum of Torture," which introduces readers to an exhibit space for devices such as corsets that crush internal organs and tweezers used to slowly pluck out a victim's hair: "For torture to be effective, the pain has to be spread out; it has to come at regular intervals, with no end in sight."

Ogawa's haunting prose may not be to everyone's taste, but readers willing to explore the murkier edges of the human psyche will not be disappointed.

New Genetic Twist: 4-Stranded DNA Lurks in Human Cells

Sixty years after scientists described the chemical code of life ? an interweaving double helix called DNA ? researchers have found four-stranded DNA is also lurking in human cells.

The odd structures are called G-quadruplexes because they form in regions of deoxyribonucleic acid (DNA) that are full of guanine, one of the DNA molecule's four building blocks, with the others being adenine, cytosine, thymine. The structure comprises four guanines held together by a type of hydrogen bonding to form a sort of squarelike shape. (The DNA molecule is itself a double strand held together by these building blocks and wrapped together like a helix.)

The new visualization of the G-quadruplex is detailed this week in the journal Nature Chemistry.

"I think this paper is important in showing directly the existence of this structure in vivo in the human genome, but it is not completely unexpected," said Hans-Joachim Lipps, of the University of Witten in Germany, who was not involved in the study. [See Images of the 4-Stranded DNA]

Scientists had shown in the past that such quadruplex DNA could form in test tubes and had even been found in the cells of ciliated protozoa, or single-celled organisms with hairlike appendages. Also there were hints of its existence in human cells, though no direct proof, Lipps said.

But scientists still didn't have concrete evidence for its existence in the human genome. In the new study, researchers, including chemist Shankar Balasubramanian, of the University of Cambridge and Cambridge Research Institute, crafted antibody proteins specifically for this type of DNA. The proteins were marked with a fluorescent chemical, so when they hooked up to areas in the human genome packed with G-quadruplexes, they lit up.

Next, they incubated the antibodies with human cells in the lab, finding these structures tended to occur in genes of cells that were rapidly dividing, a telltale feature of cancer cells. They also found a spike in quadruplexes during the s-phase of the cell cycle, or the phase when DNA replicates just before the cell divides.

As such, the researchers think the four-stranded DNA could be a target for personalized medicine in the future. If they could block these odd ducks perhaps they could stop the rapid cell division of cancer cells.

"We are seeing links between trapping the quadruplexes with molecules and the ability to stop cells dividing, which is hugely exciting," Balasubramanian said in a statement.

The finding "is certainly a technical (not scientific) breakthrough in designing antibodies sensitive enough to demonstrate this structure in vivo in the human genome," Lipps wrote.

Lipps and his colleagues had suggested previously these structures regulate basic biological mechanisms, such as the replication of DNA.

"What makes me personally very happy about this work is that it again demonstrates that mechanisms first described in ciliated protozoa hold also true for other organisms up to human, demonstrating the strength of this model organism," wrote Lipps wrote.

The team still has several questions about quadruplexes, such as how the structures operate. "One thought is that these quadruplex structures might be a bit of a nuisance during DNA replication ? like knots or tangles that form," Balasubramanian said.

Body Piercing ? what's the point?

Whether it?s to rebel against authority or a form of self expression, body piercing has been done for centuries and practiced worldwide by both men and women. So step aside 80s punk rockers and Miami Vice fans ? the sporting of earrings and nose rings dates as far back as 1500-5000 years ago! A labret is another form of body piercing that is attached to the facial lip (labrum) and dates back to Mayan and Aztec times. It was commonly associated with high status particularly among the tribal women. A more extreme body piercing practiced by Ethiopian tribes is done in the form of lip stretching adorned with a disc called a ?pelele?. Most common piercings are found in the ear lobe, nose, tongue, eyebrow or navel and adorned with jewellery such as gold hoops, gemstones, bars and even thorns (ouch!).

Ear Piercing

Practiced since ancient times, earrings represented nobility and wealth. Egyptians, Romans and Greeks wore gold hoops, precious gemstones or pendants shaped like sacred birds and asps. In Europe, between the 4th and 16th century, the popularity of earrings weaned for women as the ears and head were often covered. However, during the middle ages and 16th century, earring wear increased for men. Earrings were worn by noblemen to commoners, poets to sailors in countries such as Spain, England and France.

Nose Piercing

Nose piercing has a long history as well. There is a biblical reference in the Book of Genesis to a nose ring being given to Rebeka. In ancient Indian Sanskrit literature, the Vedas refers to Lakshmi?s piercings. Nose piercing is a common piercing practice in India, Pakistan, Nepal, South Asia and in Australian aboriginal culture and has in recent years gained popularity in Europe and North America. In Indian culture, a piercing on the left nostril is thought to make childbirth easier and is regarded as a symbol of beauty and status. Piercing of the septum or bridge can also be done although it is not as common.

For many people, piercings are a symbol of their individuality, culture and beliefs. Sai Vachan Creations Inc. specializes in unique pieces that can help you showcase your own style. For more information visit them at www.saivachan.com.

WikiCancel Is a Collection of Guides to Closing All Your Accounts

WikiCancel Is a Collection of Guides to Closing All Your AccountsSometimes the directions for closing an account on an online service are buried deep in a web site and impossible to find. WikiCancel is a wiki page that lists the cancellation procedures for popular services, subscriptions, and contracts.

WikiCancel works like any other wiki page. Click a link for a service you want to close, and WikiCancel shows you exactly how to do it. That in itself is handy, but WikiCancel also shows you how to cancel magazine subscriptions, and walks you through the closing contract services like a cell phone or internet. For contracts, it also usually includes the ways you avoid the cancellation fees when closing your account.

Of course, it's possible to get out of a cell phone contact without paying termination fees, and every once and a while a retention department will offer you a deal you can't refuse when you try to cancel. So, keep that in mind.

WikiCancel is a wiki, so if it looks like a few services are missing, feel free to add to the list to make it better. As for the bulk of your online accounts, check out the previously mentioned AccountKiller. If you've managed to forget where you even have accounts, dig through your email or old passwords to find old services you might not remember having.

Baby Boomers Create Niche Retirement Communities

North America?s Baby Boomers are the wealthiest generation alive today, and for many of them, retirement means more than not working; it means a completely new lifestyle.

As reported by Marketwatch, one of the newest markets for affluent Boomers is niche retirement homes and assisted living communities, which cater to a variety of residents? interests and affiliations.

?The typical cookie-cutter gated community is oversaturated?a lot of people just don?t want that,? Neil Schuster, founder of Lake Weir Living, a ?toy-friendly? Florida active living community that welcomes residents with jeeps, RVs, and boats, told the news source.

Some of the niche residences either currently in existence, or under development, include communities for RV aficionados, Indian-Americans, those looking for continuing college education, and Chinese-Americans, to name a few.

One retirement community, Nalcrest, located south of Orlando, Florida, was built about 50 years ago, and reserved exclusively for retired postal workers, who had been historically very poorly paid, one of the community?s management told the news source.

At Nalcrest, rents are kept artificially low at $520 per month, and the roughly 700 residents can enjoy a host of amenities, including a swimming pool, shuffleboard courts, a lake and a restaurant.

Would you consider a niche retirement community? If so, what kind of community would be your choice?

California Senate leader pushes national mental health plan ...

Darrel Steinberg is pushing for a national mental health plan.
Come for the party, stay for the lobbying. Or is that vice-versa?

State Senate leader Darrell Steinberg (D-Sacramento) is in Washington, D.C., this week to attend the inaugural festivities. But he?s mixing a little business with pleasure, looking to bend the ear of congressional leaders on mental health issues.

Motivated by the shootings in Newtown, Conn., Steinberg is urging Washington to adopt a national mental health policy. He presented his recommendations to a commission spearheaded by Vice President Joe Biden to come up with ideas about how do reduce gun violence.

Steinberg says his $10-billion plan to diagnose and treat mental illness would go a long way toward reducing mass shootings like the one at Sandy Hook Elementary School in Newtown, Conn., last year. Steinberg led the effort for Proposition 63, which generated about $1 billion a year for mental health programs in California.

The measure was funded with a 1% state surcharge on incomes of more than $1 million.

(AP) ? Will.i.am says though he's an active supporter of President Barack Obama, don't expect the Black Eyed Peas leader to become a politician.

"Nope. I like it from this angle," he said in an interview Saturday night.

Will.i.am attended an event for OurTime.org, a non-profit organization that encourages young people to vote. Several hundred people packed the Donald W. Reynolds Center for American Art and Portraiture, where John Legend, Common and T-Pain performed.

The event was just one of will.i.am's inaugural appearances: He's attending the Candle Light Inaugural Ball and Green Inaugural Ball on Sunday and will be a guest at Obama's public swearing in ceremony Monday and at the Inaugural Ball later that day.

He said it "feels good" to lend his hand to the Obama campaign and that he wants to see young voters do more.

"This is important for the youth to realize how powerful they are and to stay active and stay informed, and go out when it's time to vote. And not just for a presidential candidate, but for local government, too," he said.

Common kicked off the live performances with songs like "The People" and "Testify." He even danced and kissed a fan onstage ? and on the lips ? while Beyonce's "Party" blasted in the background.

"Just be whoever you are no matter what they say," Common said to the crowd.

The rapper also hit the stage with John Legend, who played piano as Common performed "The Light." Legend slowed the night's upbeat mood, crooning on songs like "Green Light," ''Tonight (Best You Ever Had)" and "Ordinary People."

Newly crowned Miss America 2013 Mallory Hagan, actress Sophie Bush and Arianna Huffington also attended.

Obama's campaign looks to legislative fights

WASHINGTON (AP) ? President Barack Obama's campaign racked up 332 electoral votes and helped the president win a second term. Now it hopes to score legislative victories and build the president's legacy.

Throngs of campaign volunteers and staff members gathered Sunday at a Washington hotel on the eve of Obama's Inauguration for a conference that served as both reunion and planning session for the next four years.

Recounting the success of the 2012 campaign, campaign officials vowed to use a new nonprofit called Organizing for Action to marshal support behind Obama's legislative agenda. The arrangement is unprecedented for a sitting president ? essentially transforming his re-election campaign into a nonprofit organization to back up his efforts in Congress.

"We know it is time to reform our broken immigration system, and we will get it done this year," said Jon Carson, a former White House official who will serve as OFA's executive director. "We are going to take it to them on reducing gun violence, on climate change. And we are going to take this network and finish some jobs that we started," he said, pointing to the sweeping health care law signed by Obama.

The conference, which took place as Obama was sworn into another term in a private ceremony at the White House, was the first gathering of activists who will form the backbone of the nonprofit group. The organization will be funded by grassroots donors and corporate money and function separately from the Democratic National Committee, the party's political arm.

For many volunteers and staffers, the conference and inaugural festivities also offered a chance to trade war stories and bask in the glow of Obama's re-election. Campaign manager Jim Messina walked onstage to a standing ovation, declaring to the faithful, "I see winners!"

Volunteers were asked to form local chapters of the group and take the organizing skills and technological know-how they learned from the campaign to support Obama's second term. Carson said the organization planned to make its mark quickly, previewing the upcoming fight over gun control in the aftermath of last month's deadly school shooting in Connecticut.

"There are dozens of districts in this country where Republicans have no business being proud of the fact that they're endorsed by the (National Rifle Association). And we will go to those districts, and we will run ads, and we will be on their Facebook pages," he said.

"We will go to those people in their districts who might not be too excited to hear that their member of Congress doesn't think we should pass an assault weapons ban or have background checks for everyone who wants to purchase a gun," Carson said. "We will take every lesson learned and create some new ones."

Campaign officials ticked off a series of numbers to emphasize their accomplishments ? 332 electoral votes, the registering of 1.8 million voters and more than 150 million door knocks and phone calls from volunteers. But speaker after speaker said it was more than numbers that re-elected a president.

"We are the most powerful room in America," said Sara El-Amine, the campaign's national training director. "Our legacy is not an email list. It's not a tech tool. It's not an analytics model. Our legacy is you."

Brazil judge removes "Fifty Shades of Grey" from town's shops

BRASILIA (Reuters) - A judge in Brazil's Rio de Janeiro state has confiscated copies of wildly-popular trilogy "Fifty Shades of Grey" and other erotic books from two stores, saying proprietors must seal the novels to prevent children leafing through them.

Police and judicial officials in the Rio town of Macae seized 64 books including 11 copies of the "Fifty Shades of Grey" series by British author E.L. James after the shops flouted Brazilian laws by failing to conceal erotic images and content deemed inappropriate for under-18s.

Officials will return the books within five days if the bookshop proprietors ensure they are sealed before being put back on display.

Judge Raphael Baddini de Queiroz Campos from the local family tribunal acted after finding a group of children gathered around a window display at one of the town's bookshops where erotic content was on display, the Rio de Janeiro justice service's web site said.

Ancient Earth's geochemistry: Some tectonic processes driving volcanic activity occurred 3.8 billion years ago

Jan. 18, 2013 ? Researchers still have much to learn about the volcanism that shaped our planet's early history. New evidence from a team led by Carnegie's Frances Jenner demonstrates that some of the tectonic processes driving volcanic activity, such as those taking place today, were occurring as early as 3.8 billion years ago. Their work is published in Geology.

Upwelling and melting of Earth's mantle at mid-ocean ridges, as well as the eruption of new magmas on the seafloor, drive the continual production of the oceanic crust. As the oceanic crust moves away from the mid-ocean ridges and cools it becomes denser than the underlying mantle. Over time the majority of this oceanic crust sinks back into the mantle, which can trigger further volcanic eruptions. This process is known as subduction and it takes place at plate boundaries.

Volcanic eruptions that are triggered by subduction of oceanic crust are chemically distinct from those erupting at mid-ocean ridges and oceanic island chains, such as Hawaii. The differences between the chemistry of magmas produced at each of these tectonic settings provide 'geochemical fingerprints' that can be used to try to identify the types of tectonic activity taking place early in Earth's history.

Previous geochemical studies have used similarities between modern subduction zone magmas and those erupted about 3.8 billion years ago, during the Eoarchean era, to argue that subduction-style tectonic activity was taking place early in Earth's history. But no one was able to locate any suites of volcanic rocks with compositions comparable to modern mid-ocean ridge or oceanic island magmas that were older than 3 billion years and were also free from contamination by continental crust.

Because of this missing piece of the puzzle, it has been ambiguous whether the subduction-like compositions of volcanic rocks erupted 3.8 billion years ago really were generated at subduction zones, or whether this magmatism should be attributed to other processes taking place early in Earth's history. Consequently, evidence for subduction-related tectonics earlier than 3 billion years ago has been highly debated in scientific literature.

Jenner and her team collected 3.8 billion-year-old volcanic rocks from Innersuartuut, an island in southwest Greenland, and found the samples have compositions comparable to modern oceanic islands, such as Hawaii.

"The Innersuartuut samples may represent the world's oldest recognized suite of oceanic island basalts, free from contamination by continental crust," Jenner said. "This evidence strengthens previous arguments that subduction of oceanic crust into the mantle has been taking place since at least 3.8 billion years ago."

Pocket (for Mac)

Pocket, the cross-platform bookmarking service formerly known as Read It Later, is now available to OS X?users. Like the browser extension and mobile apps, Pocket for Mac lets users save Web pages, video clips, and images to their accounts and access them at a later time.

So why would you use Pocket for Mac desktop client instead of the browser-based Pocket? The tweaked user interface features an improved icon layout and, more importantly, you can access content without an Internet connection. Previously, the only way to view your saved clips was to log into the Pocket website. Pocket for Mac may prove a highly valuable tool if you're a frequent Web reader.

Getting Started With Pocket
You're prompted to sign into your Pocket account, or create one, after you launch Pocket for Mac. You then arrive at a screen that's vertically split into two portions. The left column is your List of saved content. The larger, right area is where Pocket display content. You're given two options after logging in: "Save From Browser" and "More Ways to Save."

The former takes you to getpocket.com/add where you can install a Safari browser extension. Once the extension is installed, you can save any page to Pocket by clicking the Pocket icon located left of the browser's address bar. You can also add tags that should help you better organize content. The latter option displays the many ways you can save to Pocket via third-party apps (there are over 300 in total including the likes of Dolphin Browser HD?and Flipboard)

The Pocket for Mac Experience
Pocket for, regardless of which platform, pushes articles, images, and more to your Mac desktop. The pages, meme images, and other interesting online finds that I'd previously saved using mobile apps populated my Pocket for Mac library soon after I connected my Mac to Wi-Fi. Like the Pocket mobile apps, Pocket for Mac displays web pages (and video clips!) in a stripped down view that eliminates visual clutter and focuses on what's on hand. You can, of course, click an icon that lets you view full pages. However, video, unlike images and web pages, isn't available for offline playback. That may prove an irritant to the YouTube crowd, but you can watch your favorite saved clips when you take your Mac back online.

Pocket also includes a number of tools designed to enhance the app's usability. You can also archive, filter, search, and share content, which makes managing and distributing content a breeze.

The Wrap
Between our laptops, smart phones, tablets, and other Web-enabled devices, we read and view lots of online material?Pocket gives users away to not only save their favorite clips for reading at a later time, but manage them as well. If you're a Mac user who loves clipping and sharing the Web, consider Pocket an essential app.

3 Negotiation Tips For Women In Business

3 Negotiation Tips For Women In Business

Posted by Vicki Donlan on Thu, Jan 17, 2013 @ 10:05 AM

Every day millions of men and women around the world negotiate for what they want at home, at work, and in the community. We negotiate for goods, for services, for time, for love, and particularly, for money. Women actually negotiate more than men as they are the majority caretakers for children, as well as, elder family members. So why is it women get such a bad rap as being good at negotiation? woman negotiator

The answer is many women become uncomfortable when the negotiation as they see it is all about MONEY!

The fact is most negotiations are not about money. Money (price) just happens to be the universal starting point for a business negotiation. For example, you want to buy a new house and the one you have your eye on is $200,000 over your budget. Rather than finding out why the house is for sale, you decide not to even look at it as it is out of your price range. Stop and think about this!

If you were selling your house because you had been transferred and needed to move ASAP, you would call a real estate broker and get a fair market value for the property. The house/property would be listed for $X. However, your situation would not be listed (of course not). A buyer with the right terms and conditions might very well supercede the buyer willing to pay full price. Why? Because price in this circumstance is not the driving motivation for the sale.

Women in business have to negotiate every day with employees, vendors and customers. We have to negotiate time, terms, conditions, and, yes, MONEY. As a business coach, working with women entrepreneurs, I have witnessed time and time again my client's expertise in negotiating time and terms, but money and price often is an obstacle.

These same women successfully negotiate bedtime, nights out, chores and a multitude of other household issues, but get stymied adjudicating financial transactions.

Here are 3 tips to better negotiate every day money transactions for women in business:

(1) Listen carefully (sometimes what you want will be said between the lines)

What is actually most important to the person you are negotiating with? The more information you have the more likely you will be able to make a successful deal. Understand most people start at a price that is more than they think they can get. It seems to be built in to us to ask for the moon and settle for a star. Let the person you are negotiating with know that the price is too high by using body language - act shocked or surprised by stepping back when the price is spoken.

(2) Do your homework and know your limits - stick to them

You can't negotiate successfully if you don't really know what you want. Just as the the person you are negotiating with has an agenda so should you. What is most important to you in a successful transaction? Is it really money or is it something else? I have worked with clients who go into employee salary negotiations with such varying goals that it is no wonder they are frustrated with themselves and the negotiation process when they come out paying much more to an employee than they intended. Whose fault is that? Clearly, not the employee's fault for asking for what (s)he wants. Every negotiation needs to be entered into with an honest validation of each person's goals. Take one issue at a time and find the common denominators for both participants. There should be some areas of agreement in every negotiation - find them and communicate them. Avoid ultimatums - they rarely work in negotiations.

(3) Don't Be Afraid To Walk Away (SAY NO)

In every negotiation you have the power to walk away - use it if necessary. For example, I have a long term client who hates the proposal process in her business. She realizes she must price her service accordingly to make sure she can deliver her best work and pay her vendors what they require. Each year she has one client who consistently balks at her price and beats her down to a price that is unworkable for her business. We work on her negotiation technique before each meeting with this client. Recently, it has become clear that this client enjoys his ability to over power my client and hold her hostage. In this year's proposal, my client added an addendum: "Due to the long term relationship between our firms, we want to offer you a 15% discount to show our appreciation for your business. This year's rate of $$$$$ is effective if signed on or before 12/31/12. Proposals not signed by that date will not be considered for renewal for 2013. Thank you."

She was able to stand her ground and let the client know she was no longer interested in a no win game. BTW - the client signed - no negotiation or beat down necessary.

Successful negotiation is possible in almost every situation if you focus your attention on what the person you are negotiating with REALLY wants rather than what YOU want. Asking open-ended questions helps you get the information you need to keep the conversation directed at what outcome the other person will agree on.

So women in business PLEASE don't let money and price scare you away from negotiations. Just pretend you are negotiating with a child and allow the other person to talk and talk and talk and talk and almost wear themselves out sharing why they need or want whatever it is. Let them know they have been heard and respected and then, and only then, respond with some mutual agreement where you both can declare victory or at least begin negotiations from a place of equal value.

PFT: Vilma's lawsuit against Goodell dismissed

Philadelphia Eagles Introduce Chip KellyGetty Images

It has been presumed that new Eagles coach Chip Kelly will be using an offense in Philly that relies upon a mobile quarterback.

That?s presumption may have been premature.

As explained by Reuben Frank of CSNPhilly.com, Kelly said his system isn?t predicated on a quarterback who can run.

?[There?s] perception vs. reality,? Kelly said. ?My quarterback last year [at Oregon], Darren Thomas, who is up in the CFL, we played in 14 games, he ran for 200 yards.? Everybody is like, ?Well, you run a running offense.?? Well, look at the statistics, it?s not that.? We don?t run designed quarterback runs or we?re snapping the ball to him and then running quarterback power. . . .

?I?ve never been that way.? We?ve run zone-read concepts, man-read concepts, where it?s a mathematical game.? If there is an extra defender in the box, your quarterback can read him and by controlling him and reading him he is basically blocking him.?

Kelly explained that his approach is to make the most of the guys on the roster.

?It?s about what tools do we have in our toolbox and what tools can we use based on the players that we have,? Kelly said.? ?I think what Jim Harbaugh has done in San Francisco and Pete Carroll did in Seattle is that they identified the strengths they had in Colin Kaepernick and Russell Wilson, and they played to them.?

Kelly is right.? Good coaches devise plays and game plans aimed at maximizing the strengths and minimizing the weaknesses of their players.? Bad coaches jam the square peg of their roster into the round hole of their beloved ?system.?

Of course, an important aspect of this approach is the composition of the roster.? Specifically, which quarterbacks the Eagles will keep and which ones they?ll add.? If Kelly acquires a mobile quarterback, there will be more running.? If he keeps a drop-back quarterback like Nick Foles, there won?t be.

And Foles could be the answer, at least for now.

?He?s tough,? Kelly said of Foles.? ?I think a lot of people don?t understand how hard it is and what toughness means to the quarterback spot.? To just be able to stand in the pocket and throw the football [is tough].? We hit him as many times as we could hit him [when Oregon faces Foles at Arizona] and he just kept getting up and making plays.

?He completed a 13-yard pass left handed against us once and I remember just standing on the sideline shaking my head [saying], ?What do we have to do to stop him??? He?s a competitor, he?s accurate, so I?m excited about that.?

Kelly, in theory, could also become even more excited about the more mobile Mike Vick.? If so, Kelly needs to do so soon, because $3 million of Vick?s $15.5 million base salary in 2013 becomes fully guaranteed on February 6.

Regardless, great coaches of quarterbacks (like Andy Reid and Jim Harbaugh) get the most out of whichever quarterbacks they have.? Kelly seems to be willing to follow that same approach.? In time, we?ll find out whether he is able.
